Description:
The BS-MS Associate II, Biomarker Imaging will determine the cellular abundance and localization of biomarkers in situ using novel imaging modalities. The candidate will join a diverse team responsible for the delivery of high-quality biomarker data elucidating the mechanism of action and disease pathology in both genetic and pharmacologically induced pre-clinical models of human disease. This position will be instrumental in the group’s efforts to provide evidence of target engagement across the PTC Therapeutics pipeline with a particular interest in neurodegenerative programs. The incumbent will be responsible for communicating experimental results to his/her supervisor and working cross-functionally across multiple departments.
Primary responsibilities include:

Independently carries out grossing, orienting, and cryo-embedding research tissues
-
Maintain diagnostic viability of all specimens received and ensure correct labeling of all materials

Responsible for cryo-sectioning and thaw-mounting fresh frozen tissues
-
Perform rapid H&E or Cresyl Violet staining to quickly assess tissue coordinates and architecture
-
Prepare and maintain tissue fixatives and reagents/solutions for staining of tissue sections
-
Utilize an automated spray-coating device for high-resolution tissue derivatization and matrix applications compatible with imaging mass spectrometry
-
Conduct IHC, ICC, and IF to determine the localization of biomarkers and/or pharmacological targets

Scan and image stained slides for analysis (bright field, fluorescence and/or confocal)

Perform digital image analysis using various computer programs such as FlexImaging or ImageJ
-
Aid in non-histological experiments in support of in vitro and ex vivo biomarker quantitation for pre-clinical studies (e.g. tissue processing for quantitation of lipids, DNA oxidation, etc.)

Keep records of laboratory protocols and maintenance of equipment in an ELN

Demonstrates compliance with corporate, departmental, and job-specific requirements
Qualifications/Experience:
-
Bachelor’s degree in a scientific discipline with a minimum of 2 years of relevant experience

Skilled at cryo-sectioning and thaw mounting fresh frozen tissues

Experience with tissue and/or cell staining protocols

Proficiency with Microsoft Office

Excellent verbal and written communication and skills
-
Analytical thinker with excellent problem-solving skills and the ability to adapt to changing priorities and deadlines

Excellent planning, organization and time management skills including the ability to support and prioritize multiple projects
- Previous experience with whole-body cryo-sectioning a plus
- Working knowledge of sample preparation for imaging mass spectrometry preferred
