Description:
The Department of Drug Metabolism & Pharmacokinetics is a critical part of the Discovery Sciences organization in Janssen R&D and is committed to the delivery of high-quality clinical candidates in all Therapeutic Area (TA) discovery teams. We are currently seeking an outstanding individual to join our mechanistic in vitro ADME team as a PhD level Scientist to support the Discovery and Early Development portfolio. This individual will work closely with other DMPK team members to provide timely and seamless delivery of quality data and with other functions (Chemistry, Biology) to guide drug candidate selection. This position will be based in San Diego, CA, and may require <10% travel.
Key Responsibilities:
- New assay development to answer challenging project questions and support innovation within the DMPK department
- Design and conduct in vitro experiments to address project specific issues related to compound metabolism and interaction with enzymes/transporters
- Interpret and translate in vitro data to human predictions
- Analytical LC/MS/MS method development, bioanalysis, and troubleshooting.
- Authoring reports and supporting documentation for regulatory filings.
- Data processing and interpretation.
Qualifications/Experience:
Education:
The qualifications include a PhD in Pharmaceutical Sciences or related field with 0+ years of industry experience. Strong understanding of DMPK principles and prior experience with basic in vitro ADME techniques using cells or sub-cellular fractions, hands-on experience in LC-MS bioanalysis are required.
Experience and Skills:
Required:
- The candidate should have a strong technical understanding of DMPK principles and in vitro ADME assay design and translation to in vivo findings.
- Experience designing and executing in vitro assays to understand the mechanism of in vivo findings.
- Hands-on experience with LC-MS (ideally Sciex platform) sample analysis.
- Fluency in applying software (Excel, GraphPad Prism, Analyst) for data analysis and graphics.
- Excellent interpersonal, organizational, oral/written communication and teamwork skills.
Preferred:
- Application of PBPK modeling to translate in vitro findings to clinical predictions of PK parameters or drug-drug interactions (DDI).
- Experience with long-term hepatocyte co-culture systems such as HuREL or HepatoPac.
- Hands-on experience investigating enzyme kinetics to determine parameters such as Km, Vmax, KI, and kinact.
